    Linking urban design to sustainability : formal indicators of social urban sustainability field research in Perth, Western Australia

    The making of a livable urban community is a complex endeavor. For much of the 20th Century plannersand engineers believed that modern and rational decision-making would create successful cities. Today, political leaders across the globe are considering ways to promote sustainable development and the concepts of New Urbanism are making their way from the drawing board to the ground. While much has changed in the world, the creation of a successful street is as much of an art today as it was in the 1960s.Our work seeks to investigate 'street life' in cities as a crucial factor towards community success. What arethe components of the neighborhood and street form that contributes to the richness of street life? To answer this question we rely on the literature. The aim of the Formal Indicators of Social Urban Sustainability studyis to measure the formal components of a neighborhood and street that theorists have stated important in promoting sustainability. This paper will describe how this concept helps to bridge urban design and sustainability. It will describe the tool and show how this was applied in a comparative assessment of Joondalup and Fremantle, two urban centers in the Perth metropolitan area

    Rapid earthquake loss updating of spatially distributed systems via sampling-based bayesian inference

    Within moments following an earthquake event, observations collected from the affected area can be used to define a picture of expected losses and to provide emergency services with accurate information. A Bayesian Network framework could be used to update the prior loss estimates based on ground-motion prediction equations and fragility curves, considering various field observations (i.e., evidence). While very appealing in theory, Bayesian Networks pose many challenges when applied to real-world infrastructure systems, especially in terms of scalability. The present study explores the applicability of approximate Bayesian inference, based on Monte-Carlo Markov-Chain sampling algorithms, to a real-world network of roads and built areas where expected loss metrics pertain to the accessibility between damaged areas and hospitals in the region. Observations are gathered either from free-field stations (for updating the ground-motion field) or from structure-mounted stations (for the updating of the damage states of infrastructure components). It is found that the proposed Bayesian approach is able to process a system comprising hundreds of components with reasonable accuracy, time and computation cost. Emergency managers may readily use the updated loss distributions to make informed decisions

    Product Service System Innovation in the Smart City

    Product service systems (PSS) may usefully form part of the mix of innovations necessary to move society toward more sustainable futures. However, despite such potential, PSS implementation is highly uneven and limited. Drawing on an alternate socio-technical perspective of innovation, this paper provides fresh insights, on among other things the role of context in PSS innovation, to address this issue. Case study research is presented focusing on a use orientated PSS in an urban environment: the Copenhagen city bike scheme. The paper shows that PSS innovation is a situated complex process, shaped by actors and knowledge from other locales. It argues that further research is needed to investigate how actors interests shape PSS innovation. It recommends that institutional spaces should be provided in governance landscapes associated with urban environments to enable legitimate PSS concepts to co-evolve in light of locally articulated sustainability principles and priorities

    Direct visualization of electroporation-assisted in vivo gene delivery to tumors using intravital microscopy – spatial and time dependent distribution

    BACKGROUND: Electroporation is currently receiving much attention as a way to increase drug and DNA delivery. Recent studies demonstrated the feasibility of electrogene therapy using a range of therapeutic genes for the treatment of experimental tumors. However, the transfection efficiency of electroporation-assisted DNA delivery is still low compared to viral methods and there is a clear need to optimize this approach. In order to optimize treatment, knowledge about spatial and time dependency of gene expression following delivery is of utmost importance in order to improve gene delivery. Intravital microscopy of tumors growing in dorsal skin fold window chambers is a useful method for monitoring gene transfection, since it allows non-invasive dynamic monitoring of gene expression in tumors in a live animal. METHODS: Intravital microscopy was used to monitor real time spatial distribution of the green fluorescent protein (GFP) and time dependence of transfection efficiency in syngeneic P22 rat tumor model. DNA alone, liposome-DNA complexes and electroporation-assisted DNA delivery using two different sets of electric pulse parameters were compared. RESULTS: Electroporation-assisted DNA delivery using 8 pulses, 600 V/cm, 5 ms, 1 Hz was superior to other methods and resulted in 22% increase in fluorescence intensity in the tumors up to 6 days post-transfection, compared to the non-transfected area in granulation tissue. Functional GFP was detected within 5 h after transfection. Cells expressing GFP were detected throughout the tumor, but not in the surrounding tissue that was not exposed to electric pulses. CONCLUSIONS: Intravital microscopy was demonstrated to be a suitable method for monitoring time and spatial distribution of gene expression in experimental tumors and provided evidence that electroporation-assisted gene delivery using 8 pulses, 600 V/cm, 5 ms, 1 Hz is an effective method, resulting in early onset and homogenous distribution of gene expression in the syngeneic P22 rat tumor model

    The Tourist Experience of Heritage Urban Spaces: Valletta as a Case Study

    This article provides an understanding of how tourists experience heritage urban spaces by investigating features that influence tourist experiences most. It is framed within urban design literature which refers to three elements of urban space namely physical setting (or form), activity, and meaning. These elements are used to explore how urban spaces are experienced by tourists. Its findings are derived from an in-depth qualitative analysis of interviews with tourists to Valletta, Malta. The research suggests that the intrinsic qualities of the space are relevant to the tourist experience but what is even more relevant are the interactions of the tourist with different elements within that space, namely interactions with surroundings, interactions with others, and interactions with self/meaning. Within this broad conceptual model, the research identifies important sub-themes. Some of these reinforce the findings of existing work on tourist experiences, but others are often under-estimated or neglected

    Planning and Designing Walkable Cities: A Smart Approach

    Walking may be considered one of the most sustainable and democratic ways of travelling within a city, thus providing benefits not only to pedestrians but also to the urban environment. Besides, walking is also one of the means of transport most likely subjected to factors outside an individual\u2019s control, like social or physical abilities to walk and the presence of comfortable and safe street infrastructures and services. Therefore, improving urban conditions provided to pedestrians has positive impacts on walkability. At the same time technological solutions and innovations have the power to encourage and support people to walk by overcoming immaterial barriers due to a lack of information or boring travel and they give to decision makers the possibility to gain data to understand how and where people travel. Merging these two dimensions into a unique approach can drastically improve accessibility, attractiveness, safety, comfort and security of urban spaces. In this context, this paper aims to draw a more multifaceted context for walkability, where new technologies assume a key role for introducing new approaches to pedestrian paths planning and design and thus for enhancing this mode of transport. Indeed, by combining more traditional spatial-based and perceptual analysis of the urban environment with technological applications and social media exploitation there will be room to better support the decision on and to enhance satisfaction of walking as well as to easier plan and design more walkable cities
